The FUNNIEST performance in a film (any type of film)?

Tools

Bill Murray in Stripes
John Belushi in Animal House
Rodney Dangerfield in Caddyshack
Chevy Chase in Vacation
Zach Galifianakis in The Hangover
Kevin Spacey in American Beauty

My personal favorites:

Jack Lemmon in Some Like It Hot

George C. Scott in Dr. Strangelove

Dianne Wiest in Bullets Over Broadway

Gene Wilder in Young Frankenstein

Next to these, there are also several performances from Peter Sellers, John Cleese, Charlie Chaplin and Woody Allen, who I consider the greatest film comedians of all time.

A lot of people mention Lebowski's Walter but, though I do love him, he's not my favorite Lebowski character. I love Jesus a little more.

Also I heard in an interview with Turturro that he improvised both the ball licking and ball cleaning scenes. Mad respect to him for that.

And when I'm all alone I feel I don't wanna hide
I'll go for a more unconventional option and choose John Goodman from The Big Lebowski.

I second Peter Sellers in Dr Strangelove

Also Slim Pickens and Harvey Korman in Blazing Saddles

Dick Shawn and Kenneth Mars in The Producers

Half the cast of The Boondock Saints

Jeff Anderson in Clerks

Jason Mewes in any Kevin Smith movie

Robert Downy Jr in Tropic Thunder

Bright light. Bright light. Uh oh.
William Demarest, Raymond Walburn, Franklin Pangborn, Porter Hall, Al Bridge, Robert Grieg, Eric Blore and the rest of Preston Sturges' regular supporting players.

Skepsis93 nailed it.

I thought that might be the case but since I don't know you and it's sometimes difficult to determine tone on the internet, I figured I'd ask.

That's alright.

My avatar. May be the greatest comedic performance of all time recorded in film:

He was great, but,

> Sellers

The Brave Little Weeman Returns!
Black Dynamite - Michael Jai White

"How did you get in here?"
"...I walked in."

Pretty much anything he's in - Bill Murray

On the outside looking in.
May be the greatest comedic performance of all time recorded in film:

For once I may have to agree with you.

We've gone on holiday by mistake
Johnny Depp in Fear & Loathing In Las Vegas.

Beat me to it! Benecio Del Toro was equally awesome.
